Laser Microbeam and Optical Tweezers - Micromanipulation of Gametes and Embryos

The practical applications of contact-free laser micro-manipulation to in vitro fertilization are demonstrated. The UV laser microbeam and IR laser optical tweezers are shown in conjunction with an optical microscope. The techniques illustrated include use of laser scissors for blastomere fusion in murine 2-cell embryos, opening of the oocyte zona pellucida to facilitate sperm penetration and immobilization of sperm by tail excision. The use of tweezers is shown for trapping and dragging active sperm, holding them in the optical plane as well as performing force measurements for sperm motility. Both techniques are used as a "non-contact" option in artificial fertilization to facilitate sperm-oocyte fusion. The video shows mainly microscopic shots, but also explains some technical procedures by means of animation sequences.

Endosymbiosis between Nostoc and Geosiphon pyriforme

The incorporation of the cyanobacterium Nostoc by the Zygomycete Geosiphon is the only known example of an extant endosymbiosis between a fungus and a cyanobacterium. It is regarded as a model for the origin of plants. The fungus is also closely associated with mosses with which it can co-exist in a symiotic, mycorrhiza-like relationship, as established by recent research. In addition, its endosymbiotic partner Nostoc lives symbiotically within the thallus of mosses in this biocenosis. This video illustrates for the first time the incorporation of the cyanobacterium and its further development into a bladder. This comprehensive and high-quality educational aid was realized by means of time-lapse cinemicrography as well as electron microscopy and images of the natural habitats. An introductory sequence explains the history of discovery of the fungus. This video is designed for research and education of university level.
