Laser Microbeam and Optical Tweezers - Physical Principles. Applications in Cell Biology and Biotechnology

With the aid of laser beams (UV and IR) which can be coupled with the light path of an optical microscope, objects such as cells can be more gently manipulated without mechanical contact. Nematophagous Fungi

Nematophagous fungi use different mechanisms of capture, before killing and digesting nematodes: the endoparasites use their spores, the predatory fungi have developed special capture organs formed on hyphae, such as adhesive and constricting mycelial structures. Pleurotus infects nematodes by simple hyphae after having immobilized them with toxins. Verticillium infects eggs of cyst and root knot nematodes, also by simple hyphae. Der räuberische Pilz Monacrosporium cionopagum erbeutet Nematoden mit Hilfe von Klebfallen, die aus ein- oder mehrzelligen spezialisierten Hyphen bestehen. Wie im Film gezeigt, berührt das Opfer eine Klebfalle oft mehrfach, ehe es schließlich haftenbleibt. Der Schwerpunkt des Films liegt auf dem anschließenden Penetrationsvorgang, der in Zeitraffung dokumentiert ist und anhand von elektronenmikroskopischen Aufnahmen erläutert wird. In die erbeuteten Nematoden wachsen schließlich Ernährungshyphen ein, das Tier wird vollständig verdaut und resorbiert.

Crystallization of Evaporites in Saltworks

The saltworks of Secovlje (Portoroz, Slovenia) have been producing salt for 700 years. They can be used as a model for the study of microbiology and geochemistry of evaporite sequences of the geological past. The concentration of seawater and the crystallization of carbonates, gypsum and halite (NaCl) are documented and illustrated by graphics and microscope sequences. See films B 1839 and C 1906.

Mitteleuropa, Schleswig - Brennen von Torfkohle

The production of charcoal from peat for use in the metal industry played a significant part in the local economy around the peat-bogs on the German-danish border around Bramstedtlund until the end of the 19th century. In 1924 H. C. Davidsen was the last to use this process. Here he demonstrates again the making of grass sods, lighting the fire, placing dried peat on the fire, covering with grass sods and sand to make a kiln and, 4 weeks later, removing the cover and excavating the charcoal.

The film documents stages in the 16 1/2 hours of the course of labour for an approximately 20-year-old primiparous woman. She is attended by her mother and several other women. Trapping of Nematodes by Constricting Rings of Arthrobotrys dactyloides and Dactylaria brochopaga

Certain nematode-trapping Deuteromycetes form ring-shaped traps with a mechanical function. Touched by a nematode or stimulated artificially - e. g. by heating - the ring-cells swell and capture the nematode. The swelling is documented by slow-motion sequences, details are shown by means of electronmicrographs. The further course of infection events is shown by time-lapse sequences.

Salzbildung in Meerwassersalinen

Die geochemischen und mikrobiologischen Bedingungen in den Eindunstungsbecken sind vergleichbar mit Salzbildungen in der geologischen Vergangenheit. After the Khegebshuh mask has arrived from a neighbouring village and appeared in front of the spectators, other mask groups perform their dances, accompanied by their own instruments (xylophone, drums): the medicine society Ngang, Ndanyi from Lui, Subi from Mboh and Nkie from Lui.

On Corpus Christi the path the procession takes through the middle of the town is strewn with flowers. Various local groups are involved. Flowers, grass and branches are collected and arranged into motives. On the morning further motives are layed out using templates. Only the priest with the monstrance walks on the carpet of flowers. The film also shows the entry of the procession into the church and the culmination of the ceremony.

Herring weirs are large permeable dams made from young branches to catch herring when they migrate upriver in spring. The Schlei is a 30 km long creek where, in the middle ages, over 60 weirs were placed. Life Cycle of the Rust Fungus Puccinia graminis

Rust fungi are phylogenetically very old pathogens attacking cereals and other plants. The life cycle of Puccinia graminis involves five different spore types on two alternate hosts, barberry and cereals or grasses. It is therefore suitable as an exemple of rusts in teaching. In collaboration with John Webster, its life cycle has been documented for the first time. With the help of time lapse and cinemicrography processes such as germination, formation and discharge of spores were observed. The complicated nuclear behaviour is also shown. Graphs are used to structure, explain and summarize the topic. This video can be used to teach students at university or schools, but is also appropriate for private study.

Two men strike each with two bamboo sticks the idiophone ketuk, a thick bamboo cane with a longitudinal slit and two small sticks fastened at one end opposite to each other in prolongation of the drum. The slit drum is played in the rice-fields to keep birds away and for entertainment.
